Tubacin, without directly stabilizing microtubules, induces an increase in α-tubulin acetylation with EC50 of 2.5 μM in A549 cells. This compound inhibits HDAC6-mediated α-tubulin deacetylation, and inhibits the migration of both wild-type and HDAC6-overexpressing cells. It in combination synergistically enhances tubulin acetylation. This chemical significantly inhibits both drug-sensitive and drug–resistant MM cell growth with IC50 of 5–20 μM, and induces cell apoptosis by activation of caspases. |

Enzymhemmtests werden unter Verwendung der Reaction Biology HDAC Spectrum Plattform durchgeführt. Die HDAC1-, 2-, 4-, 5-, 6-, 7-, 8-, 9-, 10- und 11-Assays verwendeten isoliertes rekombinantes menschliches Protein; der HDAC3/NcoR2-Komplex wird für den HDAC3-Assay verwendet. Das Substrat für die HDAC1-, 2-, 3-, 6-, 10- und 11-Assays ist ein fluorogenes Peptid aus den p53-Resten 379-382 (RHKKAc); das Substrat für HDAC8 ist ein fluorogenes Diacylpeptid, das auf den Resten 379-382 von p53 (RHKAcKAc) basiert. Acetyl-Lys(trifluoracetyl)-AMC-Substrat wird für die HDAC4-, 5-, 7- und 9-Assays verwendet. Verbindungen werden in DMSO gelöst und in einem 10-Dosis-IC50-Modus mit 3-facher serieller Verdünnung ab 30 μM getestet. Die Kontrollverbindung Trichostatin A (TSA) wird in einem 10-Dosis-IC50 mit 3-facher serieller Verdünnung ab 5 μM getestet. IC50-Werte werden durch Kurvenanpassung der Dosis-Wirkungs-Steigungen extrahiert.
