SAG regulates Smo activity by binding directly to the Smo heptahelical bundle. SAG induces Smo-dependent signaling through Gli in a GRK2-dependent way. SAG also (1 nM) induces proliferation of neuronal and glial precursors without affecting the differentiation pattern of newly produced cells. |

Assays zur Hh-Signalweg-Aktivierung in Shh-LIGHT2-Zellen, einer klonalen NIH 3T3-Zelllinie, die Gli-abhängige Firefly-Luciferase und konstitutive Renilla-Luciferase-Reporter stabil integriert, werden durchgeführt. Für Studien von SAG (ein Chlorobenzothiophen-haltiger Hh-Signalweg-Agonist) und PA-SAG werden Shh-LIGHT2-Zellen bis zur Konfluenz in 96-Well-Platten kultiviert und dann mit verschiedenen Konzentrationen dieser Verbindungen in DMEM, das 0,5 % (Vol./Vol.) Rinderkälberserum enthält, behandelt.
